Взлётная масса возросла до 61 т, грузоподъёмность — до 20 т. Выпускался на Воронежском авиационном заводе (1961—1965) и Ташкентском авиационном заводе (1961—1972). Изготовлено 155 самолётов. Ан-12Б — модернизированный. Отличается усиленным центропланом, увеличенным до 19500 л запасом топлива, составом оборудования. Выпускался с 1963 года на Воронежском и Ташкентском авиационном заводе.
